Revealing <i>Candida glabrata</i> biofilm matrix proteome: global characterization and pH response

چکیده

Candida glabrata is a clinically relevant human pathogen with the ability to form high recalcitrant biofilms that contribute establishment and persistence of infection. A defining trait auto-produced matrix, which suggested have structural, virulent protective roles. Thus, elucidation matrix components, their function modulation by host environment crucial disclose role in C. pathogenesis. As major step toward this end, study aimed reveal, for first time, proteome biofilms, characterize it bioinformatic tools its environmental pH (acidic neutral). The results showed presence several pH-specific proteins (51 acidic- 206 neutral-specific) also commonly found at both conditions (236). Of note, related mannan ?-glucan metabolism, potential delivery/organization carbohydrates were but much higher quantity under neutral environment. Additionally, virulence-related proteins, including epithelial adhesins, yapsins moonlighting enzymes, among proteins. Importantly, seem non-canonical secretion pathway Pdr1 was be regulator proteome. Overall, indicates impact cues provides unique resource further functional investigation contributing identification targets development new therapies against biofilms.

Bacillus subtilis functional genomics: global characterization of the stringent response by proteome and transcriptome analysis.

The stringent response in Bacillus subtilis was characterized by using proteome and transcriptome approaches. Comparison of protein synthesis patterns of wild-type and relA mutant cells cultivated under conditions which provoke the stringent response revealed significant differences. According to their altered synthesis patterns in response to DL-norvaline, proteins were assigned to four distin...

Does the proteome encode organellar pH?

Inherent to the proteome itself, may be information that enables proteins to buffer pH at a level that promotes their own function within a specialized compartment. We observe that the distribution of computed isoelectric points in the yeast proteome matches experimentally derived organellar pH estimates across distinct subcellular compartments. This raises an interesting evolutionary question:...

Ubiquitinated proteome: ready for global?

Ubiquitin (Ub) is a small and highly conserved protein that can covalently modify protein substrates. Ubiquitination is one of the major post-translational modifications that regulate a broad spectrum of cellular functions. The advancement of mass spectrometers as well as the development of new affinity purification tools has greatly expedited proteome-wide analysis of several post-translationa...
